熒光素是一種常用的熒光素酶表達的活體成像生物發光報告子。以ATP和Mg2+作為輔因子，有氧條件下熒火蟲熒光素酶與水溶性底物反應發出一種特有的黃綠色發射光，在37℃活體內轉換為紅光。該反應通過消耗ATP來指示是否存在能量或生命的存在。
D-熒光素在生物技術領域是一種常用的試劑，尤其在活體成像方面。熒光素酶被接種在研究動物中比如大鼠，小鼠，用來標記腫瘤細胞，干細胞或傳染病。通過生物熒光成像這個模式系統中D-熒光素的注射可以用來實時，非侵入性監測疾病的進程或藥物效率。
